An international team of researchers has provided new insights into the earliest stages of protein synthesis, confirming the essential role of the nascent polypeptide-associated complex (NAC). The study, published in the journal Nature, demonstrates that NAC acts as a central chaperone, engaging with ribosomes during the translation process to guide the initial folding of nascent protein chains.

The research, involving scientists from institutions including the Medical Research Council Laboratory of Molecular Biology and Goethe University Frankfurt, utilized advanced cryo-electron microscopy. This allowed them to visualize how NAC binds near the ribosome's exit tunnel, interacting with the emerging polypeptide to prevent premature and incorrect interactions that could lead to misfolding.

Protein misfolding is linked to numerous neurodegenerative diseases, such as Alzheimer's and Parkinson's.
